Experiment: Making a Spore Solution Jar and Spore Syringe at Home
Materials Needed:
- spore print
- 300ml sterilized water in a glass jar with injection port
- Sterile gloves
- 21g sterile syringe and needle
- Alcohol swabs
- Flame or alcohol lamp
- Agar plate (for testing spore solution)
Procedure:
- Prepare the Environment: Sterilize your workspace by wiping down all surfaces with alcohol and work in a still air box if available. Wear sterile gloves to avoid contamination.
- Open the Jar: Carefully unscrew the lid of the sterilized jar under sterile conditions. Ensure that the jar remains clean and free of any contaminants.
- Add Spore Print: Using a sterile inoculation loop or a sterilized blade, scrape a small portion of the spore print into the jar. Avoid touching the inner rim of the jar to keep it sterile.
- Seal the Jar: Immediately screw the lid back on the jar to prevent contamination. Gently shake the jar to evenly disperse the spores in the water, creating a spore solution.
- Test Spore Solution: To ensure the solution is clean, remove a small amount using a 21g syringe. Wipe the injection port with an alcohol swab, then inject the spore solution onto an agar plate. Incubate the agar plate at 25-26°C to check for mycelium growth or contamination.
- Create Spore Syringe: After confirming that the spore solution is clean, sterilize a 21g syringe. Wipe the jar’s injection port with alcohol and use the syringe to draw out the desired amount of spore solution.
- Store the Solution: After use, reseal the jar and cover the injection port with micro-pore tape to ensure it stays clean. Store the jar in a fridge (2-8°C) for future use.
