Objective: To create a liquid culture for mushroom growing using a Luer-Lock syringe with needle for sterile inoculation.
Materials:
- Sterile Luer-Lock syringe with needle (2.5ml, 10ml or 20ml)
- Sterile liquid culture jar (with Self healing injection port)
- Spore syringe
- Alcohol swabs
- Still air box or flow hood
- Micro-pore tape
Procedure:
- Sterilization: Wipe the injection port of the liquid culture jar with an alcohol swab. Ensure sterile conditions by working inside a still air box or flow hood.
- Syringe Preparation: Attach the needle to your Luer-Lock syringe securely. If using a spore syringe, draw a small amount (around 1-2ml) of spore solution into the Luer-Lock syringe.
- Inoculation: Insert the syringe needle into the injection port of the liquid culture jar. Slowly inject the spore solution into the jar, ensuring you do not introduce any contaminants.
- Shake & Incubate: After inoculation, shake the liquid culture jar to evenly distribute the spores. Seal the injection port with micro-pore tape to prevent contamination. Incubate the jar at 25-27°C, shaking it daily to promote even mycelium growth.
- Observation: After 7-10 days, observe the growth of the mycelium. The culture should begin to colonize the liquid. At 50% colonization, the liquid culture is ready for further inoculations.
